Background

ILT3 (also known as LILRB4/CD85k/LIR-5) is an approximately 60 kDa transmembrane glycoprotein containing two immunoglobulin-like domains in its extracellular region and three ITIM inhibitory motifs in its intracellular region, with a secreted isoform detectable in serum. This protein is primarily expressed in myeloid cells such as monocytes and dendritic cells, as well as vascular endothelial cells. By recognizing MHC class I molecules and transmitting inhibitory signals, it negatively regulates immune cell activation and can induce the generation of CD8⁺ suppressor T cells, thereby promoting immune tolerance. Clinically, ILT3 is involved in suppressing transplant rejection, while the upregulation of its soluble form in the tumor microenvironment is associated with immune evasion, making it a potential target for immunotherapy.
